Background N(6)-methyladenosine (m6A) is one of the most abundant modified nucleosides found in some viral RNA and eukaryotic RNA including mRNA, tRNA, rRNA and lncRNA. It has been reported that m6A modification is involved in the regulation of mRNA export, stability and translation. alkB homolog 5, RNA demethylase (ALKBH5) and FTO are AlkB family proteins that function as mammalian m6A demethylases (‘erasers’). ALKBH5 catalyzes the direct removal of m6A modification on nuclear RNA and its demethylation affects RNA processing and RNA export. Whereas FTO is highly expressed in brain, ALKBH5 is highly expressed in testis and knockout of Alkbh5 gene in mice exhibits impaired male fertility.
